This made it the 18th largest of 80 barangays in City of San Pablo by population, placing it in the upper half of the city. Residents of Barangay San Diego accounted for approximately 1.78% of the total population of City of San Pablo, which stood at 285,348 in the same census year.

The barangay recorded 5,011 residents in the 2015 intercensal count and 5,068 residents in the 2020 Census — a net change of +1.1%, consistent with a slowly expanding settlement. Barangay San Diego is officially classified as urban, reflecting the density and built-up character of the area.

City of San Pablo is a city comprising 80 barangays, and serves as the governing unit directly responsible for local services in Barangay San Diego, including public health, sanitation, peace and order, and civil registration. Within the wider province of Laguna, which contains 681 barangays across its component cities and municipalities, Barangay San Diego ranks 175th by 2020 population.
